概述:该 E.coli 感受态细胞适用于高效级转化且菌落生长迅速。

基因型:F´proA+B+ lacIq ΔlacZM15/ fhuA2 Δ(lac-proAB)glnV zgb-210::Tn10 (TetR) endA1 thi-1 Δ(hsdS-mcrB)5

特性:

1)通过 lacI q 精确控制表达,从而允许克隆潜在的毒性基因

2)在琼脂糖平板上有最高的培养效率,转化 6.5 小时后即能得到可见菌落

3)挑取过夜培养的单菌落,培养 4 个小时后即进行DNA 的提取

4)可做蓝/白斑筛选

5)缺失非特异性核酸内切酶 IendA1)活性,可用于制备高质量质粒

高效级:13 X 109 cfu/μg pUC19 DNA

电转级:> 1 X 1010 cfu/μg pUC19 DNA

未被转化的细胞需要检测抗 Φ80 噬菌体感染的能力,这是检测抗 T1 噬菌体感染能力的标准检测法。此外,未转化细胞还要检测其对氨苄青霉素、氯霉素、卡那霉素和四环素的敏感性。结果显示这些未被转化的细胞抗呋喃妥因。使用 pUC 19质粒通过对 β-半乳糖基因的 α 互补分析显示:这些细胞也适宜进行蓝/白斑筛选。

注意事项:感受态细胞应贮存于–80°C,一旦融化,就不能再被冻存。–20°C贮存会显著降低其转化效率。只要温度高于–80°C,即使未被融化,细胞转化效率也会降低为零。
