Okayama Berg 用于 cDNA 克隆

概述催化双链 DNA 粘性末端的 5´-磷酸和 3´-羟基形成磷酸二酯键。

来源纯化自重组 E.coli 594su-)菌株,该菌株携带有噬菌体 λgt4 lop11 lig + Sam 7

反应条件1 X  E. coli  DNA  连接酶缓冲液 [30 mM Tris-HClpH 8.0),4 mM MgCl21 mM DTT26 μM NAD+50 μg/ml BSA]。最适反应温度为16

质保声明E. coli DNA 连接酶经过严格的质控检测,确保该产品具有最高的活性和纯度。

单位定义1 单位指在 20 μl 反应体系中,在 16 反应条件下,30 分钟能使 50% 的经 HindIII 消化的 λDNA 片段 [5 端浓度为 0.12 μM 300 μg/ml] 连接所需的酶量。

浓度10,000 units/ml

热失活65°C 20分钟。

注意事项与 rATP 作辅因子的其它连接酶不同,本酶要求NAD+(烟酰胺腺嘌呤二核苷酸)作辅因子。

 

该酶对平齐末端片段的连接效率极低,平齐末端的连接请用 T4 DNA 连接酶。

 

不要用于将 RNA 连接至 DNA
